how do you fix a pipe that leaking?

Straight line with threads coming off the main (welded), then a straight cutoff valve, then threaded PVC. It's leaking at the connection right after the valve. Tried that real thin plumber tape, dont hold the pressure. Can't see how to unscrew it, cause it would tighten the other side.
