Should my plumbing cleanout have water visible?

My toilet is not draining, when i plunge the cleanout outside overflows. the cleanout is just outside the bathroom in the yard, can i snake it and if so what direction? towards the house, the neighbors, the street? my toilet is the only fixture not draining please help - Jay
